If you have UVB lighting: yes. Non-D3 goes in the dish (your gecko self-regulates), and D3 is for dusting once a week. Without UVB: you can use D3 for everything, but the Starter Kit gives you both so you're covered if you upgrade to UVB later. Full breakdown here.
